HBM Holdings Ltd. (2142.HK) announced promising preclinical data for its AI-developed obesity drug, LET003, which showed a 76% fat mass reduction in animal models when combined with semaglutide.
"The rapid advancement of LET003 has been strongly supported by our Hu-mAtrIx™ AI platform," said Dr. Jingsong Wang, Chairman of HBM Holdings, in a statement.
In obesity models, the combination therapy decreased fat mass by 34.7% more than semaglutide alone while increasing lean mass by 5.7%, mitigating a common side effect of GLP-1 drugs. The drug, a monoclonal antibody targeting ACVR2A/2B, also showed superior dose efficiency, with a 5 mg/kg dose achieving lean mass effects comparable to a 15 mg/kg dose of competing antibody bimagrumab.
The results position HBM's antibody as a potential best-in-class therapy in the multi-billion dollar obesity market. Data from cynomolgus monkey models also showed LET003 has superior pharmacokinetic properties, suggesting it could be administered with longer dosing intervals or at lower doses than competing therapies.
The ACVR2A/2B signaling pathway is critical in regulating the body's balance of fat and muscle. While GLP-1 drugs like semaglutide are effective for weight loss, they often cause a reduction in lean muscle mass. HBM's data suggests that combining LET003 with a GLP-1 can enhance fat loss while preserving that muscle, leading to superior body composition management.
The positive data provides a key validation for HBM's Hu-mAtrIx AI platform and its drug development pipeline. Investors will now watch for the company to file for an Investigational New Drug (IND) application to begin human clinical trials, which would be the next major catalyst for the program.
